| 背景信息 | Discs large MAGUK scaffold protein 4 (DLG4), commonly known as PSD-95, is a major scaffolding protein of the postsynaptic density in glutamatergic neurons. It organizes the synaptic architecture by clustering NMDA and AMPA receptors, along with adhesion molecules like neuroligins, via its PDZ, SH3, and guanylate kinase-like domains. PSD-95 is fundamental to synaptic plasticity and transmission strength. Dysregulation of DLG4 is strongly implicated in neuropsychiatric disorders, including schizophrenia and autism spectrum disorder. Alterations in its expression or function disrupt synaptic circuitry and spine morphology, contributing to cognitive dysfunction and behavioral abnormalities. |
