| 背景信息 | INSM transcriptional repressor 1 (INSM1) is a zinc finger transcription factor that serves as a master regulator of neuroendocrine differentiation. It contains five conserved zinc-finger motifs for DNA binding and an N-terminal SNAG domain that recruits corepressors such as HDAC1/2 and KDM1A to suppress target genes, including NeuroD1 and insulin. INSM1 is essential for the development of pancreatic islets, the sympathoadrenal lineage, and neocortical progenitors. It also induces cell cycle arrest by binding to cyclin D1 and RACK1. Due to its high expression in neuroendocrine malignancies, such as small cell lung cancer and neuroblastoma, INSM1 is utilized as a sensitive diagnostic marker and a potential target for tumor-specific therapies. Its role in coordinating the transition from progenitor cells to mature endocrine cells makes it a critical factor in both development and oncology. |
