| 背景信息 | Ankyrin 2 (ANK2), also known as ankyrin-B, is a critical scaffolding protein that bridges integral membrane proteins with the spectrin-actin cytoskeleton. It is highly expressed in cardiac muscle and the brain, where it is essential for the localization and stabilization of ion channels and transporters, including Na/Ca exchangers and Na/K ATPases. In cardiomyocytes, ankyrin-B ensures the precise positioning of these transport complexes, which is vital for the electrical signaling that maintains normal heart rhythm. It also supports the stability of the neuromuscular junction in skeletal muscle and participates in polarized cell trafficking and endocytosis. Clinically, mutations in ANK2 are a well-established cause of Long QT Syndrome 4 and various cardiac arrhythmia syndromes. These conditions result from the failure of ion channels to properly localize to the plasma membrane, leading to life-threatening abnormal heart rhythms. |
