| 背景信息 | Inhibitor of DNA binding 2 (ID2) is a nuclear transcriptional regulator of the ID family characterized by a helix-loop-helix (HLH) dimerization domain. Lacking a basic DNA-binding region, ID2 cannot bind DNA directly; instead, it functions as a dominant-negative inhibitor by forming heterodimers with basic HLH (bHLH) transcription factors. This interaction prevents bHLH factors from binding to their target DNA sequences, thereby modulating critical cellular processes including growth, senescence, differentiation, and apoptosis. ID2 plays a pivotal role in lineage commitment, negatively regulating skeletal and cardiac muscle differentiation while restraining lymphoid development. Clinically, dysregulated ID2 expression is implicated in tumor biology, particularly in the development and therapy resistance of glioblastoma and Ewing sarcoma. By controlling proliferation and differentiation, ID2 contributes to oncogenesis and represents a context-dependent therapeutic target in various malignancies. |
