| 背景信息 | Glucosaminyl (N-acetyl) transferase 1 (GCNT1) is a glycosyltransferase enzyme encoded by the GCNT1 gene in humans. It catalyzes the transfer of N-acetylglucosamine (GlcNAc) in beta1-6 linkage from UDP-GlcNAc to mucin-type core 1 O-glycans, forming core 2 O-glycans. GCNT1 plays a crucial role in the biosynthesis of branched O-glycans and is essential for the formation of Gal beta 1-3(GlcNAc beta 1-6)GalNAc structures. The enzyme is localized to the Golgi apparatus and is expressed in various tissues, including the colon, pancreas, and prostate. GCNT1 is involved in the synthesis of sialyl Lewis x antigens on mucin-type glycoproteins. Overexpression of GCNT1 has been associated with the progression of several cancer types, particularly prostate cancer, where it is linked to higher levels of core 2 O-sLe(x) in PSA, PAP, and MUC1 proteins. |
