| 背景信息 | Guanylate cyclase activator 2A (GUCA2A), also known as guanylin, is a small peptide hormone primarily secreted by the intestinal epithelium. It serves as the endogenous ligand for the transmembrane receptor guanylate cyclase C (GUCY2C). Upon binding, GUCA2A stimulates the production of intracellular cGMP, which in turn regulates electrolyte and fluid secretion, maintains the mucosal barrier, and suppresses epithelial cell proliferation. This signaling axis is essential for intestinal homeostasis and the prevention of excessive fluid loss. Clinically, the loss of GUCA2A expression is a well-documented early event in colorectal tumorigenesis, where its absence promotes genomic instability and tumor progression. Conversely, overactivation of the GUCY2C pathway by guanylin-like toxins or mutations can lead to secretory diarrhea. GUCA2A is therefore recognized as a critical tumor suppressor in the colon and a key regulator of intestinal fluid dynamics. |
