| 背景信息 | Mucin 2, oligomeric mucus/gel-forming (Mucin-2) is a high molecular weight glycoprotein secreted by intestinal goblet cells that serves as the primary structural component of the protective mucus barrier in the colon. This large protein undergoes extensive O-glycosylation in the Golgi, creating bottle-brush-like domains that bind water to form a viscous gel. Mucin-2 molecules dimerize via C-terminal disulfide bonds and further oligomerize to create a two-layered barrier that protects the gut lumen from pathogens while housing the commensal microbiome. This barrier is essential for nutrient absorption and the prevention of inflammation. Dysregulation of Mucin-2, such as reduced glycosylation or sulfation, is a hallmark of ulcerative colitis, where impaired barrier integrity leads to increased susceptibility to intestinal inflammation and pathogen invasion. |
