| 背景信息 | Eukaryotic translation initiation factor 4 gamma 1 (EIF4G1) is a large scaffolding protein that serves as a core component of the eIF4F complex, which is essential for cap-dependent mRNA translation initiation. It facilitates the recruitment of mRNA to the ribosome by coordinating interactions between eIF4E, eIF4A, PABPC1, and eIF3. Through its N-terminal interaction with poly(A)-binding protein, EIF4G1 circularizes mRNA to enhance translation efficiency. It also plays a specialized role in the translation of mitochondrial oxidative phosphorylation mRNAs, supporting energy metabolism and neuronal morphogenesis. Clinically, mutations and splicing disruptions in EIF4G1 are linked to Parkinson's disease 18 and autism spectrum disorder, where deficiency impairs synaptic proteomes and cognitive functions. |
