| 背景信息 | TLE family member 2, transcriptional corepressor (TLE2) is a nuclear protein that functions as a homotetrameric corepressor to regulate gene expression through chromatin remodeling. It contains glutamine-rich, serine-proline-rich, and tryptophan-aspartate repeat domains that facilitate interactions with various transcription factors to negatively regulate DNA-templated transcription and canonical Wnt signaling. TLE2 localizes to the nucleus and focal adhesions, where it recruits epigenetic and transcriptional effector proteins to mediate gene repression. In viral contexts, it inhibits Kaposi's sarcoma-associated herpesvirus replication by repressing the replication-transcription activator. Clinically, TLE2 acts as a tumor suppressor in pancreatic ductal adenocarcinoma, where high expression levels correlate with improved patient survival and enhanced sensitivity to gemcitabine through cell cycle regulation. Conversely, its downregulation is associated with poor prognosis and the activation of oncogenic pathways such as Kras, NF-κB, and mTOR signaling. |
