| 背景信息 | Regenerating family member 3 alpha (REG3A) is a secreted C-type lectin that plays multifaceted roles in innate immunity, tissue repair, and metabolic regulation. It contains a C-type lectin-like domain with a conserved EPN motif that enables calcium-independent binding to peptidoglycans, providing bactericidal activity against Gram-positive bacteria in the intestine. REG3A expression is induced by IL-22/STAT3 signaling to support mucosal defense. In tissue repair, it promotes the proliferation of hepatocytes, keratinocytes, and gastric mucosal cells through the EXTL3/PI3K/AKT/SHP-1 signaling pathway. Furthermore, REG3A enhances glucose homeostasis and insulin sensitivity by activating AMPK. Clinically, REG3A is overexpressed in gastrointestinal cancers, such as gastric and pancreatic carcinomas, where it is linked to inflammation-driven carcinogenesis. Conversely, it exerts protective effects in inflammatory conditions like pancreatitis and metabolic disorders such as type 2 diabetes. |
