| 背景信息 | Lymphotoxin beta (LTB) is a type II transmembrane cytokine belonging to the TNF superfamily that plays a central role in immune system organization and inflammatory responses. It primarily functions by forming heterotrimers with lymphotoxin-alpha, most commonly as the LTα1β2 complex, which serves as the membrane-bound ligand for the lymphotoxin-beta receptor (LTβR). This interaction activates NF-κB and JNK signaling pathways to induce the expression of chemokines like CCL19 and CCL21, which are essential for lymphoid organogenesis and immune cell differentiation. LTB is critical for the structural maintenance of high endothelial venules and lymphocyte trafficking. Clinically, dysregulation of LTB is linked to disrupted lymphoid tissue architecture and tumorigenesis, where its overexpression has been shown to promote tumor growth and metastasis in various models. |
