| 背景信息 | E1A binding protein p300 (EP300) is a multifunctional histone acetyltransferase that plays a pivotal role in gene regulation by acetylating histones, thereby relaxing chromatin structure and facilitating transcriptional activation. As a transcriptional coactivator, p300 interacts with numerous transcription factors-including CREB, p53, and HIF1A-using specialized domains such as the bromodomain and KIX domain, and is involved in diverse cellular processes like cell growth, differentiation, and development. Beyond histones, p300 acetylates non-histone proteins, modulating their activity and stability, and is essential for normal development before and after birth. Disease associations primarily include its role as a tumor suppressor, with EP300 mutations linked to increased genome instability, altered antitumor immunity, and responsiveness to immunotherapy in various cancers. Additionally, loss-of-function mutations in EP300 cause Rubinstein-Taybi syndrome, a developmental disorder characterized by intellectual disability and distinctive physical features. |
