| 背景信息 | Forkhead box O1 (FOXO1) is a prominent transcription factor that regulates gene expression in response to insulin signaling and oxidative stress. It contains a conserved forkhead DNA-binding domain and is primarily controlled by AKT-mediated phosphorylation, which triggers its nuclear export and subsequent inactivation. In the nucleus, FOXO1 promotes gluconeogenesis and glycogenolysis in hepatocytes while inhibiting adipogenesis by repressing PPARG. It also plays a critical role in inducing apoptosis through the upregulation of FasL and Bim and controlling the cell cycle. Clinically, FOXO1 is highly relevant in metabolic and oncogenic disorders. Dysregulation is linked to type 2 diabetes, insulin resistance, and glucose intolerance. Furthermore, chromosomal translocations involving FOXO1 and PAX3 or PAX7 result in aggressive alveolar rhabdomyosarcoma, and its suppression is associated with metastasis in prostate cancer and other malignancies. |
