| 背景信息 | Discs large MAGUK scaffold protein 1 (DLG1), also known as SAP97, is a membrane-associated guanylate kinase (MAGUK) family scaffolding protein that organizes signaling complexes at specialized plasma membrane domains. It contains multiple protein-interaction domains, including three PDZ domains, an SH3 domain, and a guanylate kinase domain, which allow it to interact with ion channels, receptors, and cytoskeletal regulators. DLG1 is essential for establishing epithelial polarity, synaptogenesis, and lymphocyte activation. It specifically regulates the clustering of glutamate receptors and potassium channels at synapses and participates in ciliary protein trafficking. Clinically, DLG1 variants are associated with Crohn’s disease and congenital anomalies of the kidney and urinary tract (CAKUT), reflecting its critical role in maintaining epithelial barrier function and ciliary integrity. |
