| 背景信息 | Nuclear transcription factor, X-box binding 1 (NFX1) is a transcriptional repressor that regulates the duration of inflammatory responses by binding to the conserved X-box motif in MHC class II gene promoters, such as HLA-DRA. It contains NFX1-type zinc fingers for DNA binding and a PHD/RING domain that confers E3 ubiquitin ligase activity. NFX1 exists in multiple isoforms; NFX1-91 represses hTERT expression, while NFX1-123 stabilizes hTERT mRNA and upregulates differentiation genes like Notch1. Clinically, NFX1 is closely associated with HPV-driven oncogenesis, including cervical and penile cancers. In these contexts, the high-risk HPV16 E6 protein utilizes NFX1 to mediate the ubiquitin-dependent degradation of target proteins, leading to telomerase activation and cellular immortalization. It is also linked to epidermodysplasia verruciformis 1, reflecting its role in viral susceptibility and epithelial cell maintenance. |
