| 背景信息 | TIAM Rac1 associated GEF 1 (TIAM1) is a Rho guanine nucleotide exchange factor that specifically activates Rac1 by promoting the exchange of GDP for GTP. This activation links extracellular signals to essential cellular processes, including cytoskeletal reorganization, motility, adhesion, and polarity. Structurally, TIAM1 contains DH and PH domains necessary for its GEF activity, alongside PDZ and RBD motifs that facilitate membrane localization and interactions with partners such as talin and IRSp53. In the nervous system, TIAM1 is a critical regulator of glutamatergic synapse maturation and dendritic spine morphology. Clinically, TIAM1 is overexpressed in a wide range of malignancies, including lymphomas, neuroblastomas, and various carcinomas. Its dysregulation drives tumor invasion, metastasis, and chemoresistance through the constitutive activation of Rac1 signaling and the disruption of endothelial barriers, making it a significant therapeutic target in oncology. |
