| 背景信息 | RE1 silencing transcription factor (REST) is a master transcriptional repressor that negatively regulates neurogenesis by binding to RE1 elements in the promoters of neuronal genes. It recruits a complex of corepressors, including CoREST, mSin3A, and HDAC1/2, to orchestrate epigenetic remodeling through histone deacetylation and methylation. This silencing mechanism is essential for maintaining non-neuronal cell identity and regulating the timing of neuronal differentiation. REST also plays a role in adult neuronal integrity, oxidative stress resistance, and the regulation of insulin secretion in pancreatic beta-cells. Clinically, REST is implicated in several neurological disorders; its elevation following cerebral ischemia drives hippocampal neurodegeneration by silencing essential synaptic genes like GluA2. Furthermore, dysregulation of REST activity is associated with the pathogenesis of epilepsy and Alzheimer’s disease, where it influences neuronal viability and synaptic plasticity. |
