| 背景信息 | Cbl proto-oncogene B (CBLB) encodes CBL-B, an E3 ubiquitin-protein ligase that serves as a critical negative regulator of immune signaling and receptor tyrosine kinase (RTK) pathways. The protein contains several functional domains, including an N-terminal tyrosine kinase binding domain for substrate recognition and a RING finger domain for catalytic ubiquitin transfer. CBL-B mediates the ubiquitination and subsequent lysosomal degradation of activated RTKs, such as EGFR, while also acting as an adapter to modulate pathways like PI3K. It plays a fundamental role in maintaining peripheral tolerance by controlling T-cell costimulation and natural killer cell function. Clinically, dysregulation of CBLB is associated with various malignancies. Loss-of-function mutations or deletions promote tumorigenesis through unchecked RTK signaling, and specific mutations in the linker region are frequently observed in myeloid neoplasms such as juvenile myelomonocytic leukemia and chronic myelomonocytic leukemia. |
