| 背景信息 | Mitogen-activated protein kinase kinase kinase 1 (MAP3K1) is a multifunctional signaling node that integrates stimuli from growth factors, cytokines, and cellular stress. It possesses a C-terminal kinase domain that phosphorylates MAP2K4/7 and MAP2K1/2 to activate the JNK and ERK pathways, respectively, alongside an N-terminal PHD domain with E3 ubiquitin ligase activity. MAP3K1 regulates critical processes such as cell survival, apoptosis, and migration by scaffolding proteins like Ras, Raf-1, and TRAF2. Clinically, MAP3K1 is frequently mutated or lost in luminal breast cancer, where it plays context-dependent roles in tumor progression. Furthermore, pathogenic variants such as R186G are known to hyperactivate the Wnt4/beta-catenin pathway, leading to 46,XY disorders of sex development (DSD) through gonadal dysgenesis. |
