| 背景信息 | Tripartite motif containing 28 (TRIM28), also known as KAP1 or TIF1β, is a multi-domain protein that primarily functions as a transcriptional co-repressor. It belongs to the TRIM family and features a RING domain, two B-boxes, and a coiled-coil region, which are essential for its role in chromatin remodeling and gene regulation. TRIM28 interacts with KRAB-ZFPs (KRAB domain-containing zinc finger proteins) to repress transcription by modifying chromatin structure and recruiting histone deacetylases. Additionally, TRIM28 exhibits E3 ubiquitin ligase activity, allowing it to ubiquitinate various substrates, thereby influencing cellular signaling pathways. Its dysregulation has been implicated in several diseases, particularly cancer, where TRIM28 is associated with tumor progression and maintenance of cancer stem cell properties. Furthermore, it plays a role in the DNA damage response and apoptosis regulation. |
