| 背景信息 | Nuclear receptor subfamily 5 group A member 1 (Steroidogenic Factor 1) is a pivotal nuclear receptor and transcriptional activator that regulates the development of the adrenal glands and gonads. It binds as a monomer to specific consensus DNA sequences to drive the expression of genes essential for steroidogenesis and sexual differentiation, including CYP11A, STAR, and AMH. The activity of this receptor is modulated by phospholipid interactions and phosphorylation by kinases such as HIPK3 in response to cAMP signaling. Clinically, defects in the NR5A1 gene are well-established causes of a spectrum of disorders of sex development. These include 46,XY sex reversal with or without adrenal failure, Swyer syndrome (complete gonadal dysgenesis), and 46,XX sex reversal 4. Additionally, mutations are linked to premature ovarian failure 7 and primary adrenocortical insufficiency, reflecting its critical role in hormone production and reproductive health. |
