| 背景信息 | Signaling lymphocytic activation molecule family member 1 (SLAMF1) is a type I transmembrane receptor of the immunoglobulin superfamily, expressed on various hematopoietic cells including T and B lymphocytes, NK cells, and macrophages. It contains extracellular Ig-like domains and cytoplasmic immunoreceptor tyrosine switch motifs that recruit SH2-domain adaptors, such as SAP and EAT-2, to modulate downstream signaling through Fyn, ERK, and NF-κB. SLAMF1 acts as a self-ligand to mediate T–B cell collaboration and regulate cytokine production and phagosome maturation. It also contributes to innate immunity by sensing outer-membrane proteins of Gram-negative bacteria. Clinically, SLAMF1 serves as the primary entry receptor for the measles virus. Mutations in its signaling pathway, particularly involving the adaptor SAP, are linked to X-linked lymphoproliferative disease. It is also implicated in autoimmune thyroid disease and certain cancers, where it modulates tumor growth and immunity. |
