| 背景信息 | G protein-coupled receptor 17 (GPR17) is a class A orphan G protein-coupled receptor primarily expressed in neurons and oligodendrocyte progenitor cells within the central nervous system. It is phylogenetically related to both P2Y nucleotide and cysteinyl leukotriene receptors. GPR17 exhibits a unique mechanism of self-activation, where its extracellular loop 2 occupies the orthosteric pocket to stabilize an active conformation. Upon activation, it couples with Gi proteins to inhibit adenylyl cyclase activity and modulate potassium currents, thereby regulating the maturation of oligodendrocytes. Clinically, GPR17 is significantly upregulated following central nervous system insults, including spinal cord injury and cerebral ischemia, where it is found in proliferating microglia and macrophages. It is also implicated in the progression of glioblastoma, suggesting its potential as a therapeutic target for modulating neuroprotection and remyelination in various neurological diseases. |
