| 背景信息 | S-phase kinase associated protein 2 (SKP2) is a member of the F-box protein family that serves as the substrate recognition component of the SCF E3 ubiquitin-protein ligase complex. It plays a fundamental role in cell cycle progression by mediating the phosphorylation-dependent ubiquitination and subsequent proteasomal degradation of key negative regulators, most notably the cyclin-dependent kinase inhibitor p27Kip1. By facilitating the degradation of p27Kip1, SKP2 promotes the G1/S transition and S-phase progression. It also targets other proteins such as ORC1, CDT1, and FOXO1 for degradation. Clinically, SKP2 acts as a potent proto-oncogene and is frequently overexpressed in various malignancies. In acute myelogenous leukemia, its overexpression is an independent poor prognostic marker associated with unfavorable cytogenetics and reduced survival. Additionally, SKP2 is linked to the promotion of vascular smooth muscle cell proliferation and the development of laryngeal squamous cell carcinoma. |
