| 背景信息 | Interferon induced protein with tetratricopeptide repeats 5 (IFIT5) is a key effector of the innate immune response that functions as a sensor for viral RNA. It utilizes a unique structural architecture known as the TPR eddy to recognize single-stranded RNAs with diverse 5' phosphate structures, allowing it to distinguish viral signatures from host messenger RNA. Upon binding viral RNA, IFIT5 inhibits viral translation and modulates innate immune signaling through the activation of the NF-κB pathway. The protein is typically localized to actin-rich cellular protrusions, where it contributes to the cellular antiviral defense. Beyond its role in immunity, IFIT5 has been implicated in oncogenesis. Its upregulation is associated with enhanced cell migration, invasion, and epithelial-to-mesenchymal transition in bladder cancer and cutaneous melanoma, suggesting that it may serve as a potential biomarker or therapeutic target in specific cancer types. |
