| 背景信息 | Calcium/calmodulin dependent protein kinase II delta (CAMK2D) is a serine/threonine protein kinase that serves as a critical mediator of calcium signaling, excitation-contraction coupling, and Ca2+ homeostasis, particularly in the heart. It functions as a molecular switch, forming large multimeric holoenzymes that maintain activity following transient calcium stimuli through autophosphorylation. CAMK2D phosphorylates key targets such as HDAC4 to promote cardiac hypertrophy and titin to modulate myocyte tension. It also influences endothelial permeability via RhoA phosphorylation. Clinically, loss-of-function variants are associated with neurodevelopmental disorders characterized by intellectual disability, seizures, and autism. In the cardiovascular system, dysregulation of CAMK2D is linked to dilated cardiomyopathy, arrhythmias, and cardiac hypertrophy, often driven by disrupted calcium handling and aberrant signaling pathways. |
