| 背景信息 | Interleukin 10 receptor subunit alpha (IL10RA) is a type I transmembrane protein that serves as a critical component of the high-affinity receptor for the anti-inflammatory cytokine interleukin-10 (IL-10). Upon ligand binding, IL10RA forms a heterodimeric complex with IL10RB, triggering a conformational change that activates JAK1 and TYK2 kinases. This signaling cascade leads to the phosphorylation of STAT3, which promotes the expression of anti-inflammatory genes and inhibits proinflammatory cytokine synthesis. Additionally, IL10RA signaling suppresses autophagy and enhances myeloid progenitor survival via the PI3K/AKT pathway. The receptor is essential for maintaining mucosal homeostasis, and mutations in the IL10RA gene are well-established causes of early-onset inflammatory bowel disease 28 and increased susceptibility to chronic intestinal inflammation. |
