| 背景信息 | Ectonucleotide pyrophosphatase/phosphodiesterase 2 (ENPP2), also known as autotaxin, is a secreted glycoprotein encoded by the ENPP2 gene. It functions as both a phosphodiesterase and a phospholipase, catalyzing the production of lysophosphatidic acid (LPA) in extracellular fluids. ENPP2 plays crucial roles in various physiological processes, including embryonic development, cell motility, and angiogenesis. The protein consists of multiple domains, including two N-terminal somatomedin B-like domains, a catalytic phosphodiesterase domain, and a C-terminal nuclease-like domain. ENPP2 is involved in several pathological conditions, particularly cancer, where its expression is upregulated in various carcinomas. It stimulates tumor cell motility and promotes angiogenesis, contributing to cancer progression. Additionally, ENPP2 has been implicated in other diseases such as pulmonary fibrosis, inflammation-driven conditions like asthma and arthritis, and cardiovascular disorders. |
