| 背景信息 | Phosphodiesterase 1C (PDE1C) is a calmodulin-dependent enzyme that plays a central role in signal transduction by hydrolyzing both cAMP and cGMP. Functioning as a soluble homodimer, it regulates cyclic nucleotide levels in response to calcium signaling, thereby modulating pathways such as PKA activation. In the cardiovascular system, PDE1C influences vascular smooth muscle cell proliferation and migration, while in sensory systems, it controls the duration of cAMP signals in olfactory cilia. The enzyme is also critical for cochlear function; mutations in the PDE1C gene are well-established causes of Autosomal Dominant Deafness 74 (DFNA74), a condition characterized by progressive high-frequency hearing loss resulting from disrupted cyclic nucleotide signaling in hair cells. |
