| 背景信息 | (SCRIB) is a large LAP family scaffold protein that localizes to basolateral cell-cell junctions to organize multiprotein complexes controlling cell polarity, adhesion, and signaling. It contains multiple leucine-rich repeats and four PDZ domains that facilitate interactions with diverse partners, including beta-catenin, beta-PIX, and GIT1. Through the Scribble module, it coordinates actin dynamics and vesicle trafficking while antagonizing apical Par and Crumbs complexes to maintain basolateral identity. SCRIB is essential for regulating tissue growth and directed cell migration. Clinically, SCRIB functions as a context-dependent tumor suppressor, and its mislocalization or reduced expression is frequently linked to breast and other epithelial cancers. Additionally, perturbations in SCRIB are associated with developmental defects, particularly in neural tube formation, highlighting its fundamental role in establishing and maintaining structural organization in tissues. |
