| 背景信息 | Polypeptide N-acetylgalactosaminyltransferase 3 (GALNT3) is an enzyme encoded by the GALNT3 gene, playing a crucial role in the biosynthesis of O-linked oligosaccharides by transferring an N-acetyl-D-galactosamine residue to serine or threonine residues on proteins. This process is essential for glycosylation, impacting various biological functions, including cell signaling and protein stability. GALNT3 is particularly significant in regulating phosphate homeostasis by glycosylating fibroblast growth factor 23 (FGF23), which helps control phosphate levels in the body. Mutations in the GALNT3 gene are linked to hyperphosphatemic familial tumoral calcinosis (HFTC), a condition characterized by elevated phosphate levels and abnormal calcification due to impaired FGF23 function, leading to increased phosphate reabsorption in the kidneys. The enzyme's activity is dependent on metal ions such as manganese and calcium. |
