| 背景信息 | Glycoprotein nmb (GPNMB) is a type I transmembrane glycoprotein expressed in various cell types, including melanocytes, osteoclasts, osteoblasts, dendritic cells, macrophages, and microglia, and is structurally related to the pmel17 precursor protein. GPNMB plays diverse roles in cellular physiology: it promotes osteoblast differentiation and bone matrix mineralization, enhances fibroblast activation and proliferation, supports maturation of hematopoietic and lymphoid cells, and modulates immune responses by decreasing T lymphocyte activation. Its extracellular domain interacts with multiple receptors, such as Na+/K+-ATPase, CD44, EGFR, and VEGFR, and can bind integrins, heparin, and Syndecan-4, thereby activating downstream signaling pathways like ERK/MERK and Akt/PI3K, which influence cell survival, proliferation, and inflammation. GPNMB is also involved in tissue repair, particularly in bone regeneration, and is overexpressed in several cancers, where it may contribute to tumor progression and immune evasion. Dysregulation of GPNMB has been implicated in inflammatory and autoimmune diseases, as well as neurodegenerative disorders. |
