| 背景信息 | NODAL modulator 1 (NOMO1) is a type I transmembrane glycoprotein localized to the endoplasmic reticulum (ER) lumen. It serves as a core component of the multi-pass translocon (MPT/BOS) complex, which facilitates the insertion of multi-pass membrane proteins following their initial engagement by the SEC61 translocon. NOMO1 contains conserved immunoglobulin-like domains and forms a rod-shaped, force-bearing structure that helps maintain ER membrane spacing and sheet morphology. As one of three highly similar paralogs, it participates in protein complexes that modulate Nodal signaling during vertebrate development. Clinically, deletions of the NOMO1 gene are recurrently observed in colorectal cancer, particularly in microsatellite-stable, early-onset tumors, where they serve as a molecular marker rather than a primary driver of disease. |
