| 背景信息 | Paraoxonase 3 (PON3) is a secreted enzyme primarily synthesized in the liver and found in human plasma, where it associates with high-density lipoprotein (HDL) particles. Structurally, PON3 is a member of the paraoxonase family and exhibits strong lactonase activity, rapidly hydrolyzing lactones but not the synthetic substrates typical for PON1. Unlike PON1, PON3 is not regulated by oxidized lipids or dietary fat intake. Functionally, PON3 plays a crucial antioxidant role by inhibiting the oxidation of low-density lipoprotein (LDL), thereby protecting against oxidative stress and potentially slowing the initiation and progression of atherosclerosis. Disease associations include reduced expression in inflammatory conditions such as inflammatory bowel disease, diabetic neuropathy, and rheumatoid arthritis, while increased expression has been observed in certain tumors. These findings highlight PON3’s dual importance in lipid metabolism and disease pathophysiology, particularly in cardiovascular and inflammatory disorders. |
