| 背景信息 | Nectin cell adhesion molecule 1 (NECTIN1) is a calcium-independent cell-cell adhesion protein belonging to the immunoglobulin superfamily. It facilitates the organization of adherens and tight junctions in epithelial and endothelial cells through its three extracellular Ig-like domains. In the nervous system, NECTIN1 is vital for proper developmental patterning and synaptic contact. Beyond its physiological role in adhesion, it serves as a primary entry receptor for alphaherpesviruses, including HSV-1 and HSV-2, by binding viral glycoprotein D with high affinity. Clinically, mutations in NECTIN1 resulting in haploinsufficiency are a well-established cause of cleft lip and palate (OFC7). Its role as a viral mediator also makes it a significant factor in the pathogenesis of herpes simplex infections. |
