| 背景信息 | Protein phosphatase 2 phosphatase activator (PTPA) is a highly conserved chaperone that functions as an ATP-dependent activator for the catalytic subunit of protein phosphatase 2A (PP2A). It facilitates the acquisition of substrate specificity and enzymatic activity by stabilizing the PP2A active site and modulating metal-ion coordination. PTPA is essential for proper PP2A maturation, which in turn regulates critical signaling pathways including AKT, ERK, and MAPK. Clinically, impaired PTPA function is linked to the deregulation of PP2A, contributing to malignant transformation and therapy resistance in various cancers. In the nervous system, PTPA-mediated PP2A activity is vital for preventing the hyperphosphorylation of tau and alpha-synuclein, processes central to the pathogenesis of Alzheimer’s and Parkinson’s diseases. |
