| 背景信息 | Receptor tyrosine-protein kinase erbB-4 (ERBB4) is a member of the epidermal growth factor receptor (EGFR) family and functions as a receptor tyrosine kinase involved in various cellular processes, including growth, differentiation, and survival. ERBB4 is a single-pass type I transmembrane protein characterized by multiple cysteine-rich domains, a tyrosine kinase domain, and a PDZ-binding motif. It is activated by binding to neuregulins and other ligands, leading to receptor dimerization and autophosphorylation, which triggers downstream signaling pathways essential for cellular responses. Dysregulation of ERBB4 has been implicated in several diseases, particularly cancers such as breast cancer, lung cancer, and medulloblastoma, where mutations or altered expression levels can influence tumor progression and patient prognosis. Additionally, polymorphisms in the ERBB4 gene have been associated with neuropsychiatric disorders like schizophrenia. |
