| 背景信息 | Transcription termination factor 1 (TTF1) is a multifunctional nucleolar protein essential for the regulation of ribosomal RNA synthesis. It contains a C-terminal DNA-binding domain that specifically recognizes Sal box terminator elements, allowing it to mediate the termination of RNA polymerase I transcription. Beyond termination, TTF1 facilitates transcription initiation through the formation of chromatin loops and regulates replication fork arrest to maintain genomic stability. It interacts with various partners, including TIP5 and CSB, to coordinate chromatin remodeling and DNA damage repair. By fine-tuning rDNA transcription and cellular homeostasis, TTF1 plays a critical role in cell survival and growth. Its dysregulation is frequently observed in various malignancies, reflecting its importance in high-demand protein synthesis environments. |
