| 背景信息 | SKI2 subunit of superkiller complex (SKIC2) is a DEAD-box RNA helicase that serves as the catalytic core of the cytosolic SKI complex. It facilitates mRNA decay and quality control by unwinding RNA in the 3'-5' direction, extracting stalled mRNA from 80S ribosomes to enable ribosome recycling via Pelota-HBS1L and ABCE1. The protein contains a conserved DExH helicase domain and associates with the RNA exosome to channel substrates for degradation. SKIC2 also interacts with nuclear transcriptionally active genes through the PAF1C complex. Clinically, mutations in SKIC2 cause Trichohepatoenteric Syndrome 2, a severe autosomal recessive disorder. This condition is characterized by intractable diarrhea, woolly hair, facial dysmorphism, and liver disease, resulting from defects in mRNA deadenylation and protein folding pathways. |
