| 背景信息 | Zinc finger CCCH-type, RNA binding motif and serine/arginine rich 2 (ZRSR2) is a pre-mRNA splicing factor encoded on the X chromosome. It is characterized by CCCH-type zinc finger domains and serine/arginine-rich motifs that facilitate its interaction with the 3'-splice site of both U2-type and U12-type introns. ZRSR2 associates with the U2AF1/U2AF2 heterodimer to promote pre-spliceosome assembly in an ATP-dependent manner, playing a specialized role in the splicing of the minor (U12) spliceosome. Loss-of-function mutations in ZRSR2 are well-established drivers in myelodysplastic syndromes (MDS), where they lead to the abnormal retention of U12-type introns in critical genes, thereby impairing myeloid differentiation and contributing to leukemogenesis. |
