| 背景信息 | Discs large MAGUK scaffold protein 2 (DLG2), also known as PSD-93, is a member of the membrane-associated guanylate kinase (MAGUK) family that functions as a major scaffold protein at the postsynaptic density. It plays a critical role in organizing the molecular architecture of excitatory synapses by clustering NMDA receptors, potassium channels, and various signaling molecules. Through these interactions, DLG2 stabilizes synaptic structures and regulates the surface expression of receptors essential for glutamatergic signaling. It is prominently expressed in the cerebral cortex, hippocampus, and spinal cord, where it contributes to synaptic plasticity and the modulation of chronic pain perception. Genetic variations and deletions in the DLG2 gene are well-established risk factors for neurodevelopmental disorders, most notably autism spectrum disorder. |
