| 背景信息 | Serine/threonine kinase 11 (STK11), also known as LKB1, is a master tumor suppressor kinase that regulates cell polarity, energy metabolism, and growth. It functions as the primary upstream activator of AMP-activated protein kinase (AMPK) and several related kinases, phosphorylating them to maintain cellular homeostasis under low-energy conditions. By activating these pathways, STK11 suppresses cell proliferation, promotes autophagy, and maintains the structural integrity of epithelial tissues. Mutations that impair STK11 catalytic activity lead to uncontrolled growth and are the established cause of Peutz-Jeghers syndrome, an autosomal dominant disorder characterized by gastrointestinal hamartomatous polyps. Furthermore, loss of STK11 is a significant driver in the development of various malignancies, including lung, pancreatic, and breast cancers. |
