| 背景信息 | Interferon gamma inducible protein 16 (IFI16) is a protein encoded by the IFI16 gene in humans. It belongs to the HIN-200 family of cytokines and contains domains involved in DNA binding, transcriptional regulation, and protein-protein interactions. IFI16 localizes to the nucleoplasm and nucleoli, interacts with p53, modulates p53 function, and inhibits cell growth in the Ras/Raf signaling pathway. It plays a crucial role in sensing intracellular DNA, a hallmark of virally infected cells, and has been linked to the death of HIV-infected helper CD4 T cells by pyroptosis. IFI16 can detect various forms of DNA, including double-stranded DNA, single-stranded DNA, and quadruplex DNA, and it has been implicated in the restriction of viral replication, including HSV-1 and HIV-1. Additionally, IFI16 has been shown to induce inflammation upon TLR4 binding when extracellularly released, acting as a damage-associated molecular pattern (DAMP). |
