| 背景信息 | Thioredoxin reductase 1 (TXNRD1) is a homodimeric flavoprotein essential for maintaining cellular redox homeostasis, growth, and differentiation. It functions by reducing thioredoxin from its disulfide to its dithiol form using NADPH as an electron donor. Structurally, the enzyme features FAD and NADPH binding domains and a unique C-terminal extension containing a selenocysteine residue within a conserved Gly-Cys-SeCys-Gly motif. This configuration enables efficient electron transfer to substrates via a flexible arm, distinguishing it from prokaryotic analogs. TXNRD1 plays a critical role in regulating antioxidant defense systems and cellular signaling pathways. Dysregulation of TXNRD1 is well-established in the progression of various cancers, where redox imbalances promote tumor cell proliferation, as well as in neurodegenerative diseases and selenium-dependent pathologies. |
