| 背景信息 | ERCC excision repair 6 like, spindle assembly checkpoint helicase (ERCC6L) is a DNA helicase and member of the SWI/SNF2 family that functions as a critical tension sensor during mitosis. Utilizing its SNF2-like ATPase domain, the protein binds stretched double-stranded DNA to resolve ultra-fine DNA bridges at centromeres, telomeres, and fragile sites. This activity ensures the faithful segregation of sister chromatids before cytokinesis. ERCC6L localizes to chromatin following nuclear envelope breakdown and is also associated with ribosomal DNA. In clinical contexts, overexpression of ERCC6L is a recognized prognostic marker in several malignancies, including lung adenocarcinoma and breast cancer. It promotes tumor progression by enhancing cell proliferation, migration, and epithelial-mesenchymal transition through the activation of Wnt/beta-catenin and Notch signaling pathways. |
