| 背景信息 | Transmembrane protein 119 (TMEM119) is a type I transmembrane protein that serves as a highly specific marker for homeostatic resident microglia in the central nervous system, distinguishing them from infiltrating macrophages. It is also expressed in osteoblasts, where it promotes differentiation and bone mineralization by enhancing BMP2 signaling and interacting with RUNX2 and ATF4. In the brain, TMEM119 maintains microglial homeostasis and facilitates the phagocytosis and lysosomal degradation of amyloid-beta (Aβ) oligomers through the recruitment of LRP1. Clinical studies have established that TMEM119 expression is significantly downregulated in reactive disease-associated microglia, particularly in the vicinity of Aβ plaques in Alzheimer’s disease. This downregulation is closely linked to disease progression and neuroinflammation. Additionally, TMEM119 has been associated with malignant iris melanoma and leukoencephalopathy. |
