| 背景信息 | (Egflam) is a large extracellular matrix glycoprotein essential for the assembly and maintenance of retinal ribbon synapses. It functions as a trans-synaptic bridge, aligning the pre- and postsynaptic machinery necessary for precise visual signaling. Structurally, it contains multiple EGF-like and laminin G-like (LG) domains that mediate specific protein-protein interactions. The protein binds to presynaptic alpha-dystroglycan in a calcium-dependent manner and interacts with postsynaptic GPR179 to organize the synaptic cleft between photoreceptors and ON-bipolar cells. This organization is critical for the efficient transmission of neurotransmitters. Mutations that disrupt its function are well-established causes of congenital stationary night blindness, characterized by abnormal electroretinograms and impaired visual signal transmission. Recent studies also suggest a potential role in oncogenic extracellular matrix remodeling during glioblastoma progression. |
