| 背景信息 | (Krt18) is a low-molecular-weight type I intermediate filament protein primarily expressed in simple epithelial tissues. It forms obligate heterodimers with type II keratin 8 (KRT8) to assemble 10-nm filaments that provide essential mechanical resilience and maintain cellular integrity. The protein structure includes a central alpha-helical rod domain flanked by non-helical head and tail domains, which facilitate filament bundling and dynamic remodeling via phosphorylation. These filaments protect cells against apoptosis through PI3K/AKT signaling and modulate the cell cycle. Clinically, the protein is a well-established biomarker for epithelial-derived tumors, including hepatocellular carcinoma and breast cancer. Circulating fragments are used to monitor tumor burden and treatment response. Additionally, mutations are associated with the development of cryptogenic cirrhosis. |
