| 背景信息 | Cytidine/uridine monophosphate kinase 2 (CMPK2) is a mitochondrial enzyme within the nucleotide salvage pathway that phosphorylates monophosphates, including dUMP, dCMP, CMP, and UMP. This activity supports dNTP synthesis required for mitochondrial DNA replication and modulates macrophage inflammation by regulating mitochondrial stress and reactive oxygen species. CMPK2 plays a significant role in antiviral defense; it restricts the replication of various viruses, including flaviviruses and coronaviruses, through both kinase-dependent and independent mechanisms. It often works in conjunction with viperin to produce ddhCTP, which inhibits viral RNA polymerases. Clinically, CMPK2 is associated with autosomal recessive idiopathic basal ganglia calcification. Its dysregulation in macrophages can lead to elevated levels of pro-inflammatory cytokines such as IL-1beta and TNF-alpha, contributing to inflammatory pathologies. |
