| 背景信息 | YOD1 deubiquitinase (YOD1) is a cysteine protease belonging to the ovarian tumor (OTU) domain subfamily that plays a critical role in protein quality control. It hydrolyzes a wide range of polyubiquitin linkages, including K11, K27, K29, K33, and K48 chains, to facilitate the endoplasmic reticulum-associated degradation (ERAD) of misfolded lumenal proteins. By trimming ubiquitin chains, YOD1 enables substrate threading through the VCP/p97 pore for proteasomal degradation. Additionally, YOD1 supports macroautophagy by recruiting PLAA, UBXN6, and VCP to damaged lysosomes marked with K48-linked ubiquitin, thereby facilitating chain removal and subsequent autophagosome formation. Its enzymatic activity is essential for maintaining cellular homeostasis by managing protein turnover and organelle integrity. |
